Critical Residues of Clostridium perfringens Delta-Toxin for Oligomerization and Receptor Binding

نویسندگان

چکیده

Delta-toxin produced by Clostridium perfringens types B and C is a β-pore-forming cytotoxin. Here, using site-directed mutagenesis, we identified the amino acid residues that contribute to delta-toxin oligomerization binding. We replaced Lys-43 Ser-109 located in β-sandwich domain Arg-200 rim domain. Substitution of alanine for caused reductions both cytotoxicity oligomerization. However, exchange resulted reduction cytotoxicity. Replacement with led drastic cell Our results demonstrate plays role oligomerization, critical binding toxin.

Identification of critical residues for heterodimerization within the ligand-binding domain of retinoid X receptor.

Nuclear receptors regulate transcription by binding to specific DNA response elements as homodimers or heterodimers with the retinoid X receptors (RXRs). The identity box (I-box), a 40-amino acid region within the ligand-binding domains of RXRs and other nuclear receptors, was recently shown to determine identity in the heterodimeric interactions. Here, we dissected this region in the yeast two...

Mapping of equine lentivirus receptor 1 residues critical for equine infectious anemia virus envelope binding.

The equine lentivirus receptor 1 (ELR1), a member of the tumor necrosis factor receptor (TNFR) protein family, has been identified as a functional receptor for equine infectious anemia virus (EIAV). Toward defining the functional interactions between the EIAV SU protein (gp90) and its ELR1 receptor, we mapped the gp90 binding domain of ELR1 by a combination of binding and functional assays usin...

Structure-function analysis of heterodimer formation, oligomerization, and receptor binding of the Staphylococcus aureus bi-component toxin LukGH.

The bi-component leukocidins of Staphylococcus aureus are important virulence factors that lyse human phagocytic cells and contribute to immune evasion. The γ-hemolysins (HlgAB and HlgCB) and Panton-Valentine leukocidin (PVL or LukSF) were shown to assemble from soluble subunits into membrane-bound oligomers on the surface of target cells, creating barrel-like pore structures that lead to cell ...

Identification of HLA-DR alpha chain residues critical for binding of the toxic shock syndrome toxin superantigen

Staphylococcal toxic shock syndrome toxin 1 (TSST-1) binds to major histocompatibility complex class II molecules, and the toxin-class II complexes induce proliferation of T cells expressing V beta 2 sequences. To define the residues involved in TSST-1 binding, a set of transfectants expressing 21 HLA-DR alpha chain mutants were analyzed for their abilities to bind and present TSST-1 and to pre...
